The PI3K/AKT signaling pathway plays a critical role in numerous cellular processes, including glucose metabolism, insulin sensitivity, and cell growth, making it a crucial factor in the pathophysiology of diabetes. This paper reviews the mechanisms of the PI3K/AKT pathway and its relevance in diabetes, focusing on preclinical models and the translational potential of targeting this pathway in clinical trials. We highlight the preclinical studies that explore the therapeutic potential of PI3K/AKT modulation and evaluate ongoing clinical trials investigating its effects in diabetic patients. This review emphasizes the promise of targeting the PI3K/AKT pathway for novel diabetes treatments and underscores the need for further research to optimize therapeutic approaches.
